GDF-8 (Growth Differentiation Factor-8), also known as myostatin, is a protein encoded by the MSTN gene and a member of the TGF-β superfamily. It acts as a potent negative regulator of skeletal muscle growth, inhibiting muscle cell proliferation and differentiation. Produced and released by myocytes, it circulates in a latent form and becomes active when cleaved by proteases like BMP-1.
Active GDF-8 binds to the ActRIIB receptor, suppressing muscle growth through TGF-β signaling pathways. Loss-of-function mutations or inhibition of GDF-8 leads to dramatic increases in muscle mass—observed in “double-muscled” cattle breeds like Belgian Blue and in humans with natural mutations.
Beyond muscle, GDF-8 also influences bone density, fat metabolism, and tissue repair, with elevated levels linked to muscle wasting in conditions like heart failure, cancer, and aging. It is a key target for therapies aiming to treat muscle atrophy, sarcopenia, and metabolic disorders.
